RedHill Biopharma Receives FDA Orphan Drug Designation for YELIVA® for the Treatment of Cholangiocarcinoma

(NASDAQ:RDHL) (Tel-Aviv Stock Exchange:RDHL) (“RedHill” or the “Company”), a specialty biopharmaceutical company primarily focused on the development and commercialization of late clinical-stage, proprietary, orally-administered, small molecule drugs for gastrointestinal and inflammatory diseases and cancer, today announced that the U.S. Food and Drug Administration (FDA) has granted YELIVA® (ABC294640) Orphan Drug designation for the treatment of cholangiocarcinoma.

The Orphan Drug designation allows RedHill to benefit from various development incentives to develop YELIVA® for this indication, including tax credits for qualified clinical testing, waiver of a prescription drug user fee (PDUFA fee) upon submission of a potential marketing application and, if approved, a seven-year marketing exclusivity period for the treatment of cholangiocarcinoma.

YELIVA® is a Phase II-stage, proprietary, first-in-class, orally-administered sphingosine kinase-2 (SK2) selective inhibitor with anticancer and anti-inflammatory activities, targeting multiple oncology, inflammatory and gastrointestinal indications. By inhibiting the SK2 enzyme, YELIVA® blocks the synthesis of sphingosine 1-phosphate (S1P), a lipid signaling molecule that promotes cancer growth and pathological inflammation.

Cholangiocarcinoma (bile duct cancer) is a highly lethal malignancy for which there is a strong need for more effective systemic treatments. Approximately 8,000 people are diagnosed with intrahepatic and extrahepatic bile duct cancers annually in the U.S.1, with recent studies showing an increased incidence of cholangiocarcinoma, mainly attributed to recent advancements in diagnosis of this disease2. Surgery with complete resection remains the only curative therapy for cholangiocarcinoma, however only a minority of patients are classified as having a resectable tumor at the time of diagnosis3. Additional treatment options include radiation therapy and chemotherapy; however, the efficacy of these treatments in cholangiocarcinoma patients is also limited. Despite overall advances in the ability to diagnose and treat patients with cholangiocarcinoma, the prognosis for these relapse patients who have failed initial chemotherapy remains very poor, with an overall median survival of approximately one year4. The 5-year relative survival rates of intrahepatic and extrahepatic cholangiocarcinoma patients range between 2% to 30%, depending on the tumor type and stage at diagnosis5.

Final results from the Phase I study with YELIVA® in patients with advanced solid tumors confirmed that the study, conducted at the Medical University of South Carolina (MUSC) Hollings Cancer Center, successfully met its primary and secondary endpoints, demonstrating that the drug is well-tolerated and can be safely administered to cancer patients at doses that provide circulating drug levels that are predicted to have therapeutic activity.

Of the three patients with cholangiocarcinoma treated in the Phase I study, all of whom had prior therapy, one subject achieved a sustained partial response (Overall Survival (OS) = 20.3 months) and the other two subjects had prolonged stable disease (OS = 17.6 and 16.3 months).

RedHill plans to initiate a Phase IIa clinical study with YELIVA® in patients with advanced, unresectable, intrahepatic and extrahepatic cholangiocarcinoma in the third quarter of 2017. The single-arm study will evaluate YELIVA® as a single agent in cholangiocarcinoma patients with a primary endpoint of determining the response rate of cholangiocarcinoma to this treatment.

A Phase II study with YELIVA® for the treatment of advanced hepatocellular carcinoma (HCC) is ongoing at MUSC Hollings Cancer Center. The study is supported by a $1.8 million grant from the NCI, awarded to MUSC, which is intended to fund a broad range of studies on the feasibility of targeting sphingolipid metabolism for the treatment of a variety of solid tumor cancers, with additional support from RedHill.

A Phase Ib/II study with YELIVA® for the treatment of refractory or relapsed multiple myeloma is ongoing at Duke University Medical Center. The study is supported by a $2 million grant from the NCI Small Business Innovation Research Program (SBIR) awarded to Apogee Biotechnology Corp. (Apogee), in conjunction with Duke University, with additional support from RedHill.

A Phase I/II clinical study evaluating YELIVA® in patients with refractory/relapsed diffuse large B-cell lymphoma as well as Kaposi sarcoma patients is ongoing at the Louisiana State University Health Sciences Center. The study is supported by a grant from the NCI awarded to Apogee, with additional support from RedHill.

A Phase Ib study to evaluate YELIVA® as a radioprotectant for prevention of mucositis in head and neck cancer patients undergoing therapeutic radiotherapy is planned to be initiated in the third quarter of 2017.

A Phase II study to evaluate the efficacy of YELIVA® in patients with moderate to severe ulcerative colitis is planned to be initiated in the second half of 2017.

TGS announces further expansion of North-West African Atlantic Margin 2D seismic survey

ASKER, Norway, April 04, 2017 (GLOBE NEWSWIRE) — TGS announces a further expansion of its NWAAM multi-client library in the MSGBC Basin with plans to acquire more than 11,000 km of long-offset broadband 2D seismic data together with magnetic and gravity data, in Senegal and Gambia. This expansion is being undertaken in cooperation with Petrosen in Senegal and the Ministry of Petroleum and Energy in Gambia. The investment is being undertaken together with PGS and GeoPartners.   Acquisition of this further phase of NWAAM 2017 has commenced and continues seamlessly from Phase 1. The NWAAM 2017 seismic survey has been designed to infill, extend and complement the TGS NWAAM 2012 2D survey which helped with recent commercial discoveries in the MSGBC basin. A 12 km deep-tow streamer will enable the recording of high quality broadband 2D seismic data, which will image the pre-rift, syn-rift and post-rift plays evident in this basin.

Police to seek arrest of at least five suspects in conscript’s fatal beating

Surat Thani police are expected to seek warrants from the 45th army circle military court for the arrest of at least five suspects believed to be involved in the fatal beating of a conscript in a prison of the 45th army circle.

Pol Maj-Gen Apichart Boonsrirote, commander of Surat Thani provincial police, said the policemen in charge of the case were about to wrap the case on Tuesday night and then to seek the court's warrants for the arrest of at least five suspects.

The police team was also gathering more evidence which could implicate two more suspects, he added.

So far, about ten witnesses have been questioned by the police headed by Pol Col Wichop Kerdkliang, deputy superintendent of Surat Thani provincial police.

One of the witnesses reportedly claimed that the 22-year old victim, Pvt Yutkinant Boonniam, was assaulted on the night of March 26 at Saengpet road intersection in Muang municipal. But examination of CCTV system at the scene on that night did not show any incident.

The police inquiry team was convinced the victim was beaten up inside the prison.

Autopsy report from the forensic department of Surat Thani provincial hospital showed that Yutkinant died of kidney failure and bruises on the body were the result of assault with hard objects.

The victim's mother, Mrs Renoo told the police that she felt relieved that her son would not die in vain and that the culprits would soon be arrested.

Cabinet endorses EGAT’s new power plant in Bangpakong

The cabinet on Tuesday approved the Electricity Generating Authority of Thailand's plan to build a 1,300 megawatt power plant on the same compound of the Bangpakong electricity plant.

The 33 billion baht project will replace two electricity generating units with a combined capacity of 1,050 megawatts which were decommissioned in 2014 after they were operational for many years.

Government spokesman Lt-Gen Sansern Kaewkamnerd said that the new power plant would ensure energy security for Bangkok and its peripherals without which Bangkok may face a blackout.

Construction of the new power plant will be completed and become operation in the next two years.

The new power plant will use liquid natural gas as the main source of fuel from the Gulf supplied through the pipeline from the gas separation plant in Rayong with diesel as reserve fuel.

18 state hospitals are broke

Eighteen state hospitals are broke having accumulated debts amounting from 92 million baht up to almost 400 million baht.

The state of financial crisis of state hospitals was recently disclosed in the Facebook page of the Federation of Doctors of central and general hospitals.

The following hospitals which are considered as broke are: King Rama III hospital, 354 million baht in debt; Saraburi hospital, 321 million baht; Uttradit hospital, 277 million baht; Sakhon Nakhon hospital, 225 million baht; Surat Thani hospital, 219 million baht; Khon Kaen hospital, 214 million baht; Chao Phraya Yommaraj hospital, 199 million baht; Hua Hin hospital, 194 million baht; Wachira Phuket hospital, 185 million baht; Phayao hospital, 164 million baht; Phichit hospital, 156 million baht; Songkhla hospital, 138 million baht; Hat Yai hospital, 133 million baht; Nakhon Ping hospital, 131 million baht; King Rama II hospital, 116 million baht; Phra Phuttabaat hospital, 115 million baht; Prachuab Khiri Khan hospital, 102 million baht and Sukhothai hospital, 92 million baht.

Public Health Minister Dr Piyasakol Sakonsattayatorn admitted that state hospitals had been experiencing financial crisis for quite a long time and the situation is now critical.

However, he assured that the government would did not sit idly by and was trying to solve the problem.

He blamed the financial crisis facing the 18 state hospitals on poor management and insufficient funding from the universal health coverage programme.
